Overview

The **hemagglutinin head domain** is the membrane-distal, globular region of the influenza virus hemagglutinin (HA) glycoprotein, comprising the receptor-binding subdomain and, in Influenza A, the vestigial esterase subdomain[1][3][4][6][7]. It is primarily responsible for binding to sialic acid–containing receptors on the surface of host epithelial cells, enabling viral attachment and entry[1][4][6]. The head domain (mainly within HA1) is also the principal target for virus-neutralizing antibodies, making it a critical determinant of host immunity and vaccine efficacy[1][2][4][8]. It is characterized by rapid antigenic evolution—which facilitates evasion of host immune responses—and contains well-defined antigenic sites that are frequently updated in seasonal influenza vaccines[8]. The HA head domain is structurally distinct from the stalk (or stem) domain and is a key focus of both therapeutic monoclonal antibodies and next-generation vaccine strategies[2][4][8].
