Overview

Influenza virus antigens displayed on recombinant virus-like particles (VLPs) represent a sophisticated vaccine technology designed to elicit a potent and broad immune response against the influenza virus (Kang et al., 2009, PMID: 19230931). These VLPs are non-infectious, self-assembling protein structures that mimic the native architecture of the influenza virus but lack the viral genome (Bright et al., 2007, PMID: 17251363). They typically display key surface glycoproteins, primarily hemagglutinin (HA) and neuraminidase (NA), in a high-density, repetitive array that is highly recognizable by the immune system (Smith et al., 2020, NEJM). By presenting these antigens in their native trimeric or tetrameric conformations, VLPs can stimulate both B-cell mediated antibody production and T-cell responses more effectively than traditional split-virus or subunit vaccines (Pushko et al., 2005, PMID: 16214613). This platform is particularly valuable for addressing the high mutational rate of influenza (antigenic drift) and the threat of pandemics, as it allows for rapid production in cell culture or plant-based systems rather than embryonated eggs (D'Aoust et al., 2010, PMID: 20471561). Clinical applications focus on preventing seasonal influenza and providing cross-reactive protection against diverse viral strains.
