Overview

Inhibition of angiogenesis" refers to the therapeutic strategy aimed at preventing the formation of new blood vessels from pre-existing vasculature. This approach is primarily used in cancer treatment because tumors require new vasculature for sustained growth and metastasis. Drugs that inhibit angiogenesis typically act by blocking key molecules such as vascular endothelial growth factor (VEGF) or its receptors on endothelial cells. These agents can slow tumor progression by depriving cancerous tissue of oxygen and nutrients supplied by newly formed vessels. While effective in certain cancers and other diseases characterized by abnormal neovascularization—such as age-related macular degeneration—antiangiogenic therapies are associated with notable side effects including hypertension, impaired wound healing, bleeding risks, proteinuria, fatigue, skin changes, and rare but serious gastrointestinal perforations[1][2][3][4].

Blockade of vascular endothelial growth factor (VEGF) binding to its receptor on endothelial cells. Inhibition of intracellular signaling pathways downstream from proangiogenic receptors. Interference with paracrine signaling between tumor and stromal cells. Immunomodulation with antiangiogenic effects.
