cIAP1 is a multifunctional regulator at the intersection of cell death, survival, inflammation, and immunity. It functions as an E3 ubiquitin ligase and regulates apoptosis through ubiquitination pathways, notably by ubiquitinating RIPK1. It is involved in NF-κB signaling, MAPK pathway activation, innate immunity, and stress adaptation. Overexpression or dysregulation of cIAP1 has been observed in various cancers, contributing to tumor progression and resistance to therapy. It also plays a critical role in pancreatic β-cell survival during endoplasmic reticulum stress. As such, it represents a potential therapeutic target for cancer and other diseases.
IAP antagonists promote degradation of cIAP1, leading to apoptosis in cancer cells and modulation of inflammatory signaling.
