Overview

INCA1 (Inhibitor of cyclin-dependent kinase interacting with cyclin A1) is a highly conserved protein that binds to cyclin A1–CDK2 complexes and inhibits CDK2 kinase activity, resulting in delayed S-phase progression and suppression of cell proliferation[3]. It plays a critical role in regulating the cell cycle during spermatogenesis, acting as both a substrate and binding partner of cyclin A1–CDK2, with high expression in testicular tissue[2][6][3]. INCA1 is also implicated in tumor suppression: its loss leads to increased CDK2 activity, aberrant cell cycle progression, and impaired leukemia development[3]. Its antiproliferative effects are further linked to its interaction with other proteins like ING5[3]. INCA1 is predominantly localized in the cytoplasm and nuclear body[3]. No approved drugs target this molecule, and it is not currently considered a classic therapeutic target such as an enzyme, receptor, or transporter[3][7].
