Overview

Inhibitor of DNA binding protein 3 (ID3) is a member of the ID family of helix-loop-helix (HLH) transcription factors. ID3 lacks a DNA-binding domain and functions predominantly as a dominant-negative regulator of basic HLH transcription factors by dimerizing with them, thereby inhibiting their DNA binding and activity. ID3 plays a critical role in regulating cell growth, differentiation, and development by modulating gene expression. It is implicated in numerous physiological processes and is found to be dysregulated in multiple cancers, where it can contribute to tumorigenesis, metastasis, and resistance to cell death. Its unique regulatory mechanisms and crucial involvement in cell fate make it an attractive but challenging molecular target in cancer and regenerative medicine research.
