Overview

The "inhibitor of growth family, X-linked" (INGX) is a non-canonical or erroneous designation; all credible sources and genomic nomenclature point to five recognized ING family genes. Most references to "ING1-like" or "INGX" correspond incorrectly or ambiguously to ING2 or a general ING family member, not a distinct, X-linked protein. Inhibitor of growth family member 2 (ING2) is a chromatin-binding tumor suppressor protein in the ING family. It is characterized by a plant homeodomain (PHD), interacts with histone H3K4 trimethyl marks, and associates with the HDAC-SIN3A complex to regulate transcription, chromatin structure, DNA repair, and cell cycle control. ING2 promotes cellular senescence and apoptosis, and loss-of-function mutations or reduced expression are linked to increased genomic instability and various cancers. ING2 also localizes to mitochondria, suggesting broader roles in cell metabolism and integrity. The designation "INGX," "ING1-like," or "ING1-like tumor suppressor protein" is incorrect or outdated and should be replaced by "inhibitor of growth family member 2 (ING2)" for canonical nomenclature.

Mechanism of action
