Overview

The Inhibitor of nuclear factor kappa-B kinase (IKK) complex is a multi-protein enzyme assembly that acts as the master regulator of the NF-κB signaling pathway, which is essential for immune and inflammatory responses (PMID: 10485677). The complex typically consists of two catalytic subunits, IKKα (CHUK) and IKKβ (IKBKB), and a regulatory subunit, IKKγ (NEMO). Upon activation by stimuli such as pro-inflammatory cytokines or pathogens, the IKK complex phosphorylates IκB proteins, leading to their degradation and the subsequent nuclear translocation of NF-κB transcription factors. This pathway is frequently hyperactivated in various cancers and chronic inflammatory diseases, making the IKK complex a high-interest therapeutic target for drug development (PMID: 22460726). However, clinical progress has been limited by the complex's vital role in innate immunity and cell survival, which leads to significant safety concerns such as severe immunosuppression and hepatotoxicity. Most current IKK inhibitors are small molecules targeting the ATP-binding site of the IKKβ subunit, though few have successfully transitioned to late-stage clinical use.
