Overview

The Inhibitor of nuclear factor kappa-B kinase subunit alpha and beta (IKKα/β) complex is a critical enzymatic component of the canonical and non-canonical NF-κB signaling pathways (UniProt: P78527, O14920). These kinases are responsible for the phosphorylation of inhibitory IκB proteins, which triggers their proteasomal degradation and allows NF-κB transcription factors to enter the nucleus. Once in the nucleus, NF-κB regulates the expression of genes involved in the immune response, inflammation, cell proliferation, and survival (PMID: 11544344). In many pathological states, such as rheumatoid arthritis and various malignancies, IKKα/β becomes overactive, leading to chronic inflammation and tumor progression (PMID: 21620916). Pharmacological targeting of IKKα/β aims to suppress these pathways using small-molecule inhibitors to treat inflammatory disorders and sensitize cancer cells to chemotherapy. Despite its therapeutic potential, the development of IKK inhibitors has been hindered by significant safety concerns, including severe immunosuppression and liver toxicity, due to the essential role of NF-κB in maintaining normal cellular homeostasis and host defense mechanisms (PMID: 21620916).

Mechanism of action

Inhibition of IκB kinase catalytic activity, preventing the phosphorylation and subsequent proteasomal degradation of IκB proteins, which blocks the nuclear translocation and transcriptional activity of NF-κB (PMID: 11544344).
