IKKβ is a serine/threonine protein kinase and a key regulatory subunit within the IκB kinase (IKK) complex. It is essential for the canonical NF-κB signaling pathway, activated by diverse stimuli like inflammatory cytokines, microbial products, and stress signals. Upon activation, IKKβ phosphorylates IκBs, leading to their degradation and subsequent NF-κB translocation into the nucleus to activate genes involved in immune response, inflammation, cell survival, proliferation, and apoptosis. Inhibition of IKKβ is explored as a therapeutic strategy for inflammatory diseases and cancer.
Inhibition of IKKβ prevents phosphorylation of IκBs, leading to suppression of NF-κB activation and downstream inflammatory gene expression.
