Overview

Inhibitory Killer-cell Immunoglobulin-like Receptors (iKIRs) are a family of receptors expressed on NK cells and some T cells that recognize MHC class I molecules. iKIRs deliver inhibitory signals, preventing NK cells from killing healthy cells. The interaction between iKIRs and MHC class I molecules is critical for maintaining self-tolerance and regulating immune responses. Polymorphisms in KIR genes and their ligands contribute to individual differences in immune responses and disease susceptibility.

Mechanism of action

Phosphorylation of ITIMs recruits phosphatases (SHP-1/SHP-2) that dephosphorylate signaling proteins, inhibiting NK cell activation.
