Innate and adaptive immune receptors modulated by Alhydrogel refer to the collective molecular network through which aluminum hydroxide adjuvants stimulate the immune system. The primary mechanism involves the activation of the NLRP3 inflammasome, a multiprotein complex that triggers the maturation of pro-inflammatory cytokines like IL-1β and IL-18 (Eisenbarth et al., 2008, Nature). Alhydrogel also induces the release of endogenous danger signals, such as uric acid and host DNA, which serve as ligands for various innate pattern recognition receptors (Kool et al., 2008, J Exp Med). Furthermore, Alhydrogel interacts directly with dendritic cell membranes via lipid sorting to initiate signaling independent of traditional surface receptors (Flach et al., 2011, Nat Med). These innate signals ultimately modulate adaptive immune receptors, such as MHC class II and the T-cell receptor, to promote a robust Th2-biased antibody response (Marrack et al., 2009, Nat Rev Immunol). While highly effective for increasing vaccine potency, this target group represents a broad physiological response rather than a single drug-receptor interaction.
