Overview

Innate immune cell recruitment and activation refers to the coordinated physiological process where primary defense cells, such as neutrophils, monocytes, and natural killer (NK) cells, migrate toward sites of tissue injury or infection. This process is driven by complex signaling involving chemokines, selectins, and integrins that facilitate the rolling, adhesion, and transmigration of leukocytes across the vascular endothelium [1, 2]. Upon reaching the target tissue, these cells are activated through pattern recognition receptors (PRRs) like Toll-like receptors (TLRs), triggering the release of pro-inflammatory cytokines and the initiation of phagocytosis [3]. In oncology, therapeutic strategies often aim to enhance this process to stimulate anti-tumor immunity, whereas in autoimmune or inflammatory conditions, the goal is often to inhibit excessive recruitment to prevent host tissue damage [4, 5]. Pharmacological intervention can occur at various stages, using agents such as TLR agonists to boost activation or chemokine receptor antagonists to block cell trafficking [6]. Consequently, while it is a vital host defense mechanism, its dysregulation is a hallmark of many chronic diseases and a major focus of modern immunopharmacology.
