Innate immune cells and local tissue refers to the collective group of non-specific immune cells—such as macrophages, neutrophils, dendritic cells, and natural killer cells—and the anatomical environment they inhabit (Janeway et al., 2001). This system acts as the body's primary defense mechanism, recognizing and responding to pathogens or injury through the release of cytokines and the recruitment of further immune effectors (Medzhitov, 2008). In various diseases, including chronic inflammation and cancer, the interaction between these innate cells and the local tissue microenvironment becomes dysregulated, often promoting pathological states like fibrosis or tumor growth. While many therapeutic strategies aim to modulate this environment by targeting specific molecular pathways, such as Toll-like receptor (TLR) signaling or cytokine receptors, the term itself represents a broad physiological category rather than a single druggable molecular target (Maizels et al., 2012). Consequently, it is not classified as a canonical therapeutic target in pharmacological databases, which instead focus on specific proteins or genes. The coordination between these cells and the local tissue is essential for maintaining homeostasis and facilitating the transition to adaptive immunity.
