INAVA (innate immunity activator protein), encoded by the C1orf106 gene, is a protein involved in both innate immune signaling and epithelial barrier integrity. It is expressed in intestinal and peripheral myeloid cells and intestinal epithelial cells[1][2]. INAVA acts as a scaffold in pattern recognition receptor (PRR)-initiated signaling, interacting with 14-3-3τ, NOD2, RIP2, and IRAK1 to promote MAPK and NF-κB pathway activation and cytokine secretion, and to enhance bacterial clearance via reactive oxygen/nitrogen species and autophagy[1]. In epithelial cells, INAVA/C1orf106 interacts with cytohesin-1 and members of the SCF ubiquitin ligase complex to regulate cytohesin-1 degradation, thereby controlling ARF6 activity and adherens junction stability. Risk variants in INAVA/C1orf106 associated with IBD decrease protein expression or stability, resulting in impaired cytokine response, bacterial clearance, and epithelial barrier integrity[1][2][3].
