Inosine monophosphate dehydrogenase (IMPDH) is a key enzyme in the de novo biosynthesis of guanine nucleotides. It catalyzes the rate-limiting step: the conversion of inosine 5'-monophosphate (IMP) to xanthosine 5'-monophosphate (XMP), with concomitant reduction of NAD+ to NADH. This reaction is essential for maintaining adequate pools of guanine nucleotides, which are critical for DNA and RNA synthesis, signal transduction, and energy transfer. IMPDH is an established drug target for immunosuppressive, anticancer, and antiviral therapies. Inhibitors targeting this enzyme disrupt guanine nucleotide synthesis, thereby affecting rapidly dividing cells such as lymphocytes or cancer cells.
