Overview

Inositol monophosphatase 1 (IMPA1) is a magnesium-dependent phosphatase enzyme that catalyzes the dephosphorylation of myo-inositol monophosphate to generate free myo-inositol, which is a crucial precursor for the biosynthesis of phosphatidylinositol and polyphosphoinositides, key components of cell signaling pathways[3][4][10]. IMPA1 activity is inhibited by lithium and other small molecules. This inhibition is believed to underlie part of the therapeutic mechanism of mood stabilizers, particularly in the treatment of bipolar disorder, by modulating phosphatidylinositol-mediated signaling pathways[1][3][9]. IMPA1 is widely expressed, especially in the brain, and participates in the maintenance of inositol homeostasis and the regulation of downstream signaling and cellular functions. Disruption of IMPA1 activity can lead to alterations in autophagy, apoptosis, and other cellular processes[3].
