Inositol transport into erythrocyte-containing parasites refers to the process by which inositol is taken up from the host red blood cell (erythrocyte) environment by intracellular parasites such as Plasmodium species (malaria parasites) and Babesia. This transport is essential for parasite survival and proliferation during their intraerythrocytic stages. The parasite relies on de novo synthesis as well as salvage pathways to obtain myo-inositol, which is used for phosphoinositide metabolism—a key process in membrane formation and signaling. Solutes like inositol must cross several barriers: first entering the erythrocyte via its plasma membrane, then traversing the parasitophorous vacuole membrane (PVM), and finally crossing the parasite plasma membrane (PPM). Manipulating inositol availability can impact parasitemia. For example, treatment with highly charged compounds like inositol hexaphosphate dramatically reduces infection rates by Babesia microti both in vivo and ex vivo, suggesting that interfering with this pathway could be a viable therapeutic strategy.
