Insect carboxylesterases (CCEs) are a diverse family of α/β-hydrolase enzymes that play critical roles in the physiological development and environmental adaptation of insects. They are primarily involved in the hydrolysis of endogenous esters, such as juvenile hormones and pheromones, as well as the detoxification of exogenous xenobiotics, including plant allelochemicals and synthetic insecticides (1.2.1, 1.4.2). In the field of public health and agriculture, these enzymes are significant as both the primary targets of organophosphate and carbamate insecticides and as major drivers of metabolic resistance (1.3.2, 1.3.5). Resistance often arises through gene amplification or point mutations that enhance the enzyme's ability to sequester or rapidly hydrolyze pesticide molecules before they reach their neurological targets (1.2.2, 1.3.1). Understanding the structural and functional diversity of insect esterases is essential for developing novel pest control strategies and managing the spread of vector-borne diseases like malaria and dengue (1.3.3, 1.4.5).
