The insect cuticle is a complex, multi-layered extracellular matrix that functions as the exoskeleton, providing essential structural support, protection against environmental stressors, and a primary barrier against pathogens [1, 2]. It consists of chitin microfibrils embedded in a diverse matrix of cuticular proteins, which undergo sclerotization (cross-linking) to achieve the necessary rigidity for movement and defense [3]. While not a single molecular receptor, the cuticle is a major focus of pharmacological intervention in pest control and vector management, specifically through the disruption of its synthesis and maintenance [4]. Insect Growth Regulators (IGRs), such as the benzoylurea class (e.g., lufenuron), act by inhibiting chitin synthase enzymes, leading to defective cuticle formation and mortality during the molting process [5, 6]. Furthermore, the physicochemical properties of the cuticle, particularly the waxy epicuticle, represent a significant barrier to the penetration of contact insecticides, making cuticular components vital considerations in drug delivery and resistance research [7, 8].
Interference with cuticle development occurs primarily through the inhibition of chitin synthesis and polymerization, disruption of the hormonal signaling (ecdysteroids) that triggers molting, or inhibition of the sclerotization processes required for hardening the exoskeleton [4, 5, 6].
