Overview

Insect heteromeric olfactory receptor complexes are the primary sensory units responsible for detecting volatile chemical cues in insects, playing a vital role in survival and reproduction (Larsson et al., 2004, Nature). These complexes are composed of a variable, odor-specific odorant receptor (OR) subunit and a highly conserved, ubiquitous co-receptor known as Orco (Butterwick et al., 2018, Nature). Unlike mammalian olfactory receptors, which are G protein-coupled receptors, these insect complexes function as heteromeric ligand-gated ion channels with an inverted membrane topology (Sato et al., 2008, Nature). They are essential for host-seeking behaviors in disease vectors, such as mosquitoes that transmit malaria and Zika virus, as well as for locating food sources in agricultural pests. Because Orco is highly conserved across nearly all insect orders, it serves as a versatile target for the development of novel insect repellents and attractants (Jones et al., 2011, PNAS). Small molecules like VUAA1 act as allosteric agonists that can overstimulate the olfactory system, while other compounds act as antagonists to block odor detection, effectively blinding the insect to its environment. Targeting these complexes offers a species-specific approach to controlling insect populations and reducing the transmission of vector-borne diseases (Wicher et al., 2008, Nature).
