The Insulin-like Growth Factor 1 Receptor (IGF-1R) is a transmembrane receptor tyrosine kinase that mediates the effects of IGF-1, IGF-2 and insulin. Upon ligand binding, the receptor undergoes autophosphorylation, activating downstream signaling cascades such as the PI3K/Akt pathway, promoting cell survival, proliferation, metabolism regulation, neuroprotection, and neuroregeneration. Dysregulation of IGF-1R is implicated in various human cancers, making it an important target for anti-cancer drug development.
