Monoclonal antibodies act as antagonists by binding to the extracellular alpha-subunit of the receptor, preventing ligand binding and inducing receptor internalization, while small molecule inhibitors compete with ATP at the intracellular beta-subunit tyrosine kinase domain to block downstream signaling cascades [1, 6].
