Insulin-like growth factor 1 receptor (IGF-1R) messenger RNA is the genetic template responsible for the synthesis of the IGF-1R protein, a key mediator of cell growth, transformation, and survival (PubMed: 12467225). In many cancers, the overexpression of IGF-1R mRNA leads to an abundance of the receptor, which promotes tumor progression and resistance to apoptosis (PubMed: 10491401). Targeting the mRNA directly using antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs) offers a method to downregulate the entire IGF-1R signaling pathway at its source. This approach is particularly relevant in conditions like glioblastoma and prostate cancer, where traditional inhibitors have faced challenges (ClinicalTrials.gov: NCT02507583). However, therapeutic intervention must be carefully managed to avoid interfering with the closely related insulin receptor, which could lead to metabolic complications such as hyperglycemia.
Antisense oligonucleotides and RNA interference (RNAi) molecules bind to the IGF-1R mRNA sequence, leading to its degradation via RNase H or the RNA-induced silencing complex (RISC), thereby preventing the translation of the IGF-1R protein.
