Overview

Insulin-like growth factor 2 antisense RNA (IGF2-AS) is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the IGF2 gene locus on chromosome 11p15.5 (NCBI Gene ID: 51214). It serves as a key regulator of genomic imprinting and gene expression, particularly within the IGF2/H19 cluster, which is essential for normal growth and development (PubMed: 25633037). In various malignancies, including hepatocellular carcinoma and colorectal cancer, IGF2-AS is often upregulated and acts as an oncogenic driver by promoting cell proliferation, epithelial-mesenchymal transition (EMT), and resistance to apoptosis (PubMed: 30106434). It frequently functions as a competitive endogenous RNA (ceRNA), sequestering microRNAs to prevent them from silencing oncogenic targets. Due to its specific overexpression in tumors and its role in driving disease progression, IGF2-AS is considered a promising therapeutic target and a potential diagnostic biomarker. Experimental strategies targeting IGF2-AS, such as antisense oligonucleotides (ASOs), are currently being explored to inhibit its function and sensitize cancer cells to conventional therapies (PubMed: 31431359).
