Insulin-like growth factor-binding protein 1 (IGFBP-1) is a secreted protein that binds insulin-like growth factors I and II (IGF-1 and IGF-2) in circulation, thereby regulating their half-life and bioavailability and modulating their interactions with cellular receptors. IGFBP-1 is encoded by the IGFBP1 gene, is also known as placental protein 12, and possesses an IGFBP domain as well as a thyroglobulin type-I domain. It plays a critical role in various cellular processes including migration, proliferation, differentiation, and apoptosis, both by modulating IGF action and through IGF-independent interactions with integrins via its RGD motif. IGFBP-1 is implicated in several physiological and pathological states, including cancer, cardiovascular disease, metabolic syndrome, and pregnancy-related disorders[4][5][6].
