Insulin-like growth factor binding protein 2 (IGFBP2) regulates the actions of insulin-like growth factors IGF-I and IGF-II. It modulates IGF bioavailability, distribution, and activity in various physiological and pathological processes. IGFBP2 binds both IGF-I and IGF-II with high affinity, especially IGF-II. It controls cell proliferation, survival, differentiation via regulation of type I/II receptor signaling. It also acts as a ligand itself by interacting with cell surface integrins. Altered expression or function has been implicated in various diseases including cancer progression/metastasis, metabolic disorders, autoimmune diseases, and developmental abnormalities due to its central role in regulating cell growth signals.
