Overview

Insulin-like growth factor-binding protein 5 (IGFBP-5) is a secreted protein that regulates the actions of IGF-I and IGF-II by high-affinity binding, modulating IGF availability to their receptors, and thereby influencing cell growth, differentiation, proliferation, migration, and apoptosis[1][3][4][5][11]. IGFBP-5 is the predominant IGF-binding protein in bone and plays a key role in skeletal development and maintenance[11]. Besides extracellular effects, IGFBP-5 can enter the nucleus and act as a transcriptional coregulator, modulating gene expression independently of IGFs[13]. Dysregulation of IGFBP-5 has been implicated in cancer progression, metabolic diseases, and cardiovascular disorders due to its central role in IGF signaling modulation[4][11][13].
