Microbial membrane proteins and structures comprise a heterogeneous group of biomolecules responsible for mediating a range of functions essential for microbial survival, virulence, and host interaction. These proteins are primarily embedded in the lipid bilayer of microbes and can be classified as integral (span the membrane), peripheral (associate transiently), or lipid-anchored (covalently connected to membrane lipids). Structural motifs include α-helical bundles (most common in cytoplasmic membranes) and β-barrel architectures (predominantly in bacterial outer membranes). Functionally, microbial membrane proteins regulate transport (nutrients, ions), facilitate signal transduction and adhesion, participate in enzymatic reactions, and can mediate antibiotic resistance. Due to their essential roles and exposed location, microbial membrane proteins represent prime targets for antibiotic development and immunotherapy, but their diversity and rapid evolution pose considerable therapeutic challenges.
