Overview

Integrator complex subunit 11 (INTS11) is an RNA endonuclease and a key catalytic component of the multi-protein Integrator complex, which regulates the 3′-end processing and transcriptional termination of noncoding RNAs and a subset of protein-coding genes by RNA polymerase II. INTS11 belongs to the metallo-β-lactamase superfamily and forms a stable, functionally essential complex with Integrator complex subunit 9 (INTS9) through their C-terminal domains. The Integrator complex acts as a molecular checkpoint during transcription, promoting termination and degradation of nascent RNA when elongation is unfavorable. Disruption of INTS11 function leads to transcriptional readthrough, misprocessing of snRNAs, and global dysregulation of gene expression. INTS11 has been implicated in neurodevelopmental disease, and research continues into its role in cancer and other transcription-mediated disorders.
