Integrator complex subunit 14 (INTS14) is one of the 14 core structural proteins comprising the Integrator complex, which assembles at the C-terminal domain of RNA polymerase II. INTS14, together with INTS13 and INTS10, forms a stable nucleic acid-binding module within the Integrator, displaying affinity for DNA and RNA hairpins, although with no high substrate specificity. The module acts as an accessory factor crucial for efficient 3′-end processing of spliceosomal snRNAs and also plays a role in the termination of transcription by RNA polymerase II. Structurally, INTS14 features a von Willebrand type A (VWA) domain, a β-barrel, and an α-helical domain, and interacts tightly with INTS13, forming a physically interlinked heterodimer. While INTS14 itself has not been directly implicated in disease, it is required for spermatogenesis in model organisms and participates in broader chromatin and transcriptional regulatory mechanisms[1][3].
