Overview

Integrator complex subunit 2 (INTS2) is an essential component of the **Integrator complex**, a large multiprotein assembly associated with the C-terminal domain of RNA polymerase II. INTS2, together with other core subunits (e.g., INTS1 and INTS7), forms the structural backbone of the complex. The Integrator complex mediates the 3′-end processing of small nuclear RNAs (snRNAs) and plays a critical regulatory role in **transcriptional attenuation and termination** at protein-coding and non-coding RNA genes, largely by coupling RNA cleavage/nuclease activity and protein phosphatase 2A (PP2A)–dependent dephosphorylation of the transcriptional machinery. INTS2, while not possessing catalytic activity itself, is crucial for the structural assembly and correct targeting of the Integrator and associated enzymatic modules. Functional studies indicate that INTS2 is necessary for proper cell fate decisions during neural development by ensuring the maintenance of neural progenitor differentiation and preventing dedifferentiation. Alterations in the Integrator complex, including INTS2 deficiency, are associated with misregulation of gene expression in disease contexts, but INTS2 itself is not currently a recognized drug target or clinical biomarker[1][2][3].
