Overview

Integrator complex subunit 3 (INTS3) is a multifunctional protein that plays critical roles in both the maintenance of genome stability and in transcriptional regulation. As part of the SOSS complex, INTS3 participates in the cellular DNA damage response by promoting the repair of double-strand DNA breaks through homologous recombination and facilitating activation of ATM-dependent checkpoint signaling[1][2]. It scaffolds other SOSS components, including single-stranded DNA-binding proteins, and stabilizes the complex at DNA damage sites[1][2]. INTS3 is also a component of the Integrator complex, which regulates transcription termination by RNA polymerase II: it helps in processing small nuclear RNAs, enhancer RNAs, and additional noncoding and coding transcripts by facilitating termination in the promoter-proximal region[1][3]. Mutations or deficiency in INTS3 have been linked to genomic instability and cancer susceptibility[1].
