Overview

Integrator complex subunit 4 (INTS4) is a scaffold protein that is an essential component of the Integrator complex, a multi-protein complex associated with RNA polymerase II. INTS4 is required for the assembly of the Integrator cleavage module together with INTS9 and INTS11, which is involved in the endonucleolytic 3′-end processing of small nuclear RNAs (UsnRNAs) and enhancer RNAs. In mammalian cells, INTS4 plays a critical role in maintaining Cajal body and histone locus body integrity, necessary for proper RNA biogenesis. INTS4 physically interacts with INTS9/11 through its N-terminal HEAT repeats and C-terminal β-sheet-rich region. Loss of INTS4 disrupts snRNA processing and nuclear body assembly, affecting global RNA maturation pathways. While essential for RNA biology, INTS4 is not considered a current therapeutic target nor is it known to be implicated directly in human disease[1][2][3].
