Overview

Integrator complex subunit 5 is a protein component of the Integrator complex, a large multi-protein assembly required for the regulation of transcription mediated by RNA polymerase II. The Integrator complex is essential for premature transcription termination and RNA 3'-end processing, particularly of small nuclear RNAs, and is implicated in global regulation of gene expression. INTS5, also referred to as Integrator subunit 5, forms a stable heterodimer with INTS8, constituting the so-called 'shoulder module' of the complex, which is structurally important for assembling and stabilizing the Integrator complex scaffold. While INTS5 itself has no direct enzymatic activity, its presence is critical for the structural integrity, assembly, and potentially the locus-specific recruitment of the entire Integrator complex to chromatin. Disruption of Integrator subunits, including INTS5, affects gene expression programs and has been associated with developmental defects, cancer, and neurodevelopmental disease.
