Integrator complex subunit 8 (INTS8) is a protein encoded by the INTS8 gene and is a core component of the Integrator complex, which is essential for the 3′ end processing of small nuclear RNAs and transcriptional termination of RNA polymerase II transcripts. INTS8 acts as a multifunctional regulator of gene expression by recruiting protein phosphatase 2A (PP2A) to modulate transcriptional pause release and maintain transcriptional fidelity. Genetic alterations or dysregulation of INTS8 are implicated in neurodevelopmental disorders and several cancer types, where its overexpression is often linked to tumor progression, poor prognosis, and changes in the tumor microenvironment. Current studies highlight its emerging role as a diagnostic or therapeutic target, particularly in liver and gastrointestinal cancers.
