Overview

Integrin alpha-9 beta-1 is a heterodimeric cell surface receptor composed of the alpha-9 (ITGA9) and beta-1 (ITGB1) subunits, functioning as a member of the integrin family of cell adhesion molecules[1][2][8]. It mediates cell–cell and cell–extracellular matrix interactions by binding various ligands such as tenascin-C, VCAM1, osteopontin, ADAM family proteins, and others[1][8]. Integrin alpha-9 beta-1 is broadly expressed in human tissues and plays crucial roles in processes like cell migration, lymphangiogenesis, angiogenesis, and tissue development, as well as pathologies such as inflammation, cancer progression, autoimmune disorders, and nerve injury[2][3]. Intracellularly, its signaling includes activation of Src kinases, inducible nitric oxide synthase (iNOS), FAK, Erk, and Rac1, integrating cytoskeletal remodeling and migration cues[2][4]. Recent research highlights α9β1 as a potential therapeutic target in a range of diseases, but clinically validated drugs targeting this integrin remain under investigation[3].
