Overview

Integrin alpha FG-GAP repeat-containing protein 1 (ITFG1), also known as T-cell immunomodulatory protein (TIP), is a protein that plays a significant role in the regulation of the immune system and cellular signaling (UniProt, Q8TB96). It is characterized by its FG-GAP repeats, which are typically found in the alpha subunits of integrins, suggesting a role in cell adhesion and extracellular matrix interactions (NCBI Gene, 81533). ITFG1 is known to modulate T-cell activation and the production of inflammatory cytokines, making it a key player in immune homeostasis (PubMed, PMID: 16139227). In oncology, ITFG1 is frequently upregulated and has been linked to tumor growth, invasion, and the epithelial-mesenchymal transition (EMT), particularly through its influence on the TGF-beta signaling pathway (PubMed, PMID: 32854157). As a therapeutic target, ITFG1 is being explored for its potential to enhance anti-tumor immune responses or inhibit metastatic progression (PubMed, PMID: 35645478). Research indicates that silencing ITFG1 can inhibit the proliferation and migration of various cancer cells, including those in gastric and breast cancers (PubMed, PMID: 32854157). Despite its potential, there are currently no approved drugs or clinical-stage candidates specifically targeting ITFG1. Future therapeutic strategies may involve monoclonal antibodies or RNA-based interventions to modulate its activity in the tumor microenvironment.
