Integrin alpha-IIb beta3 (GPIIb/IIIa) is a heterodimeric transmembrane receptor complex found predominantly on the surface of platelets and megakaryocytes. It plays a central role in platelet aggregation, hemostasis, and thrombosis. In its resting state, it does not bind ligands; upon platelet activation, it undergoes conformational changes that increase its affinity for soluble ligands such as fibrinogen and von Willebrand factor. Ligand binding leads to integrin clustering and outside-in signaling, which triggers further platelet activation processes. GPIIb/IIIa is targeted by several antiplatelet drugs used primarily during percutaneous coronary interventions.
