Integrin beta-1-binding protein 1 (ITGB1BP1, commonly known as ICAP1) is an intracellular protein encoded by the ITGB1BP1 gene and acts as a key regulator of integrin-mediated cell-matrix interactions. ITGB1BP1 binds specifically to the cytoplasmic tail of beta-1 integrin (ITGB1), preventing its activation by competing with other activators such as talin. This interaction influences cell adhesion, spreading, migration, proliferation, bone mineralization, and angiogenesis. ITGB1BP1 modulates focal adhesion site dynamics, regulates Rho family GTPase activity during cell-matrix adhesion, and serves as a negative regulator of angiogenesis. Multiple isoforms exist due to alternative splicing, with only the longer phosphoprotein isoform interacting with ITGB1. Diseases associated with ITGB1BP1 include cerebral cavernous malformations and immunodeficiency, while it plays functional roles in bone and blood vessel development. Currently, it is not the direct target of approved drugs and is not classified as a clinical biomarker.
