Overview

Integrin beta-1-binding protein 2 (ITGB1BP2), also known as Melusin and CHORDC3, is a co-chaperone protein characterized by two cysteine and histidine-rich (CHORD) domains and multiple protein–protein interaction motifs, including PXXP and YXXI/P sequences[4]. It primarily interacts with the beta-1 integrin subunit in a cation-dependent manner, excluding calcium[6]. ITGB1BP2 is expressed in cardiac and skeletal muscle and participates in muscle cell maturation, organization, and possibly mechanotransduction associated with integrin signaling[5][6]. Functional data in mouse and human indicate involvement in muscle development, cell differentiation, and adaptation to mechanical stress[3][5]. While not a canonical drug target like a receptor or enzyme, its role in muscle and cardiovascular biology suggests indirect relevance in disease mechanisms, especially myocardial adaptation to stress and muscle plasticity[3][5]. Currently, there are no known drugs or clinical biomarkers that directly target ITGB1BP2, nor reported safety concerns specific to its modulation.
