Overview

Integrin beta-3 (ITGB3) is an integral membrane protein that combines with specific alpha subunits (notably alpha-IIb and alpha-V) to form the functional integrin receptors αIIbβ3 (found in platelets) and αVβ3 (expressed in various cell types)[2][3][5]. These integrins mediate cell adhesion and bidirectional signal transduction between the extracellular matrix and the cell interior, most prominently facilitating platelet aggregation and thrombus formation by binding ligands containing RGD and related sequences, such as fibrinogen, vitronectin, and fibronectin[2][3][6]. Drugs targeting integrin beta-3, such as eptifibatide and abciximab, are used clinically to inhibit platelet aggregation and prevent thrombotic cardiovascular events, but can cause bleeding complications by impairing normal hemostatic functions[3][5]. Genetic polymorphisms and immune responses against β3 integrin are associated with several pathological conditions, including thrombosis, endometriosis, and immune-mediated platelet disorders like fetal/neonatal alloimmune thrombocytopenia[1][5][6].
