Target intelligence / Profile preview
Integrin beta-3 (ITGB3), commonly referred to as CD61, is a transmembrane glycoprotein that functions as a receptor subunit within the integrin family (UniProt P05106). It forms heterodimeric complexes with either the alpha-IIb subunit (forming alpha-IIb/beta-3) primarily on platelets or the alpha-V subunit (forming alpha-V/beta-3) on various cell types, including mesenchymal stem cells (MSCs), endothelial cells, and osteoclasts. On MSCs, ITGB3 is essential for mediating interactions with the extracellular matrix, which facilitates cell migration, homing to damaged tissues, and survival (PubMed: 25600112). In the context of disease, ITGB3 is a key player in thrombosis, tumor-induced angiogenesis, and bone remodeling (PubMed: 12759929). Pharmacological targeting of ITGB3 is well-established in cardiovascular medicine through the use of glycoprotein IIb/IIIa inhibitors to prevent blood clots. However, its broad expression and critical role in hemostasis present challenges, particularly regarding the risk of hemorrhage and the need for precise therapeutic windows.
Antagonism of the integrin receptor to prevent binding of ligands such as fibrinogen, von Willebrand factor, and vitronectin, thereby inhibiting cell adhesion and aggregation (DrugBank: DB00054).
