Overview

Integrin beta-4 (ITGB4) is a transmembrane glycoprotein that functions as a subunit of the integrin family of cell adhesion receptors. It exclusively pairs with the alpha 6 subunit to form the α6β4 integrin heterodimer. This heterodimer plays a critical structural role in hemidesmosomes, anchoring epithelial cells to the underlying basement membrane. Beyond structural support, integrin β4 mediates signal transduction from the extracellular matrix into intracellular pathways, influencing cell proliferation, migration, and invasion. Mutations in ITGB4 cause epidermolysis bullosa with pyloric atresia, while overexpression or altered function contributes to tumorigenesis.
