Overview

Integrin subunit beta 1 (ITGB1) mRNA is the transcript encoding the beta-1 subunit of the integrin family, which are critical transmembrane receptors that facilitate cell-extracellular matrix (ECM) and cell-cell adhesion. These receptors function as heterodimers, pairing with various alpha subunits to integrate mechanical and chemical signals from the microenvironment into the cell, thereby regulating survival, proliferation, and migration (UniProt P05556). In many pathological states, particularly oncology, ITGB1 mRNA is significantly upregulated, driving processes such as epithelial-mesenchymal transition (EMT), tumor invasion, and metastasis (PubMed: 31434017). Furthermore, high levels of ITGB1 expression are associated with resistance to chemotherapy and radiotherapy in multiple cancer types. Targeting the ITGB1 mRNA transcript using RNA interference (RNAi) or antisense oligonucleotides (ASOs) represents a therapeutic strategy to silence the gene before protein synthesis occurs. Preclinical studies have demonstrated that reducing ITGB1 mRNA levels can effectively inhibit tumor progression and sensitize malignant cells to standard treatments (PubMed: 25666443). However, the development of such therapies must address the challenge of tissue-specific delivery to avoid interfering with the essential roles of integrins in normal tissue homeostasis and vascular integrity.

Mechanism of action

Degradation of the ITGB1 mRNA transcript via RNA interference (RNAi) or RNase H-mediated cleavage, which prevents the translation and subsequent surface expression of the Integrin beta-1 protein.
