Overview

Integrin subunit beta 7 (ITGB7) is a transmembrane protein that forms heterodimers with alpha integrin subunits, primarily α4 (CD49d) or αE (CD103). The α4β7 integrin interacts with MAdCAM-1 on endothelial cells to direct lymphocyte homing to gut-associated lymphoid tissue. The αEβ7 integrin interacts with E-cadherin on epithelial cells to mediate retention of lymphocytes within epithelial tissues. ITGB7 plays a critical role in leukocyte adhesion, migration, and immune surveillance at mucosal surfaces, and is a therapeutic target for inflammatory bowel disease and other immune-mediated disorders.

Mechanism of action

Inhibition of ITGB7 prevents lymphocyte binding to MAdCAM-1 and/or E-cadherin, thereby blocking lymphocyte trafficking to mucosal tissues and reducing inflammation.
