Interactor of HORMAD1 protein 1 (IHO1) is a coiled-coil domain-containing protein conserved across vertebrates and primarily expressed in testis tissue where it plays a critical role in meiotic recombination[2][4][9]. It forms homotetramers through a parallel four-stranded coiled-coil region and scaffolds the assembly of the MEI4–REC114–IHO1 (MCD) recombinosome complex on chromatin during meiosis[4][8]. IHO1 interacts directly with chromosome axis protein HORMAD1 to seed DSB machinery clusters, thereby enabling DNA double-strand break formation necessary for correct homologous chromosome pairing[2][3][6]. Disruption of IHO1 function or expression leads to meiotic failure and infertility but is not known to be modulated by drugs. As a cancer/testis antigen, IHO1 has potential as a biomarker for tumor subtypes with aberrant testis-like gene expression profiles[9].
