Overview

Intercellular Adhesion Molecule 1 (ICAM-1), Lymphocyte Function-associated Antigen 1 (LFA-1), and the beta-2 and beta-3 integrin families are critical cell surface glycoproteins that mediate cellular adhesion and signaling. ICAM-1 (CD54) is a member of the immunoglobulin superfamily that serves as a ligand for the integrin LFA-1 (alpha-L/beta-2), a member of the beta-2 integrin family (PMID: 15123770). This interaction is fundamental for the stable arrest and transmigration of leukocytes across the vascular endothelium during inflammatory responses (UniProt: P05362). Beta-2 integrins (CD18) are essential for innate and adaptive immunity, while beta-3 integrins (CD61), including alpha-IIb/beta-3 and alpha-v/beta-3, are vital for platelet aggregation and vascular remodeling (StatPearls: Integrin Inhibitors). Dysregulation of these molecules is linked to chronic inflammatory conditions, autoimmune disorders, and thrombotic events. Therapeutic strategies targeting these proteins include monoclonal antibodies and small molecules designed to block adhesion, such as Lifitegrast for dry eye disease and Abciximab for preventing blood clots during coronary interventions (PubChem: CID 16140448).
