Overview

The intercellular lipid lamellae of the stratum corneum (SC) represent the primary permeability barrier of the mammalian skin, located between the corneocytes in the outermost layer of the epidermis (StatPearls: Physiology, Stratum Corneum). These lamellae are composed of a unique mixture of ceramides, free fatty acids, and cholesterol organized into highly ordered crystalline structures (PubMed: The stratum corneum: structure and function in health and disease). Their primary biological function is to prevent excessive transepidermal water loss (TEWL) and to block the entry of environmental allergens, pathogens, and toxins (Journal of Investigative Dermatology: Skin barrier lipids).
