Target intelligence / Profile preview
The intercellular lipid lamellae of the stratum corneum (SC) represent the primary permeability barrier of the mammalian skin, located between the corneocytes in the outermost layer of the epidermis (StatPearls: Physiology, Stratum Corneum). These lamellae are composed of a unique mixture of ceramides, free fatty acids, and cholesterol organized into highly ordered crystalline structures (PubMed: The stratum corneum: structure and function in health and disease). Their primary biological function is to prevent excessive transepidermal water loss (TEWL) and to block the entry of environmental allergens, pathogens, and toxins (Journal of Investigative Dermatology: Skin barrier lipids). In various dermatological conditions such as atopic dermatitis and psoriasis, the composition and organization of these lipids are significantly altered, leading to barrier dysfunction and increased sensitivity (Journal of Lipid Research: Lipid composition and organization in the stratum corneum). Therapeutic interventions often target these lamellae through the application of physiological lipid mixtures to restore barrier integrity or via chemical enhancers that temporarily fluidize the lipids to facilitate drug delivery. Understanding the phase behavior and molecular arrangement of these lipids is crucial for developing effective topical treatments and transdermal delivery systems.
Restoration of the skin's permeability barrier through the replenishment of essential lipids and the creation of an occlusive layer to prevent water loss (StatPearls: Physiology, Stratum Corneum).
