Interferon alpha-inducible protein 27-like protein 2 (IFI27L2) is a member of the interferon-stimulated gene (ISG) family, closely related to other proteins such as IFI27 and IFI6, and is encoded by the IFI27L2 gene (also known as FAM14A, ISG12B, or TLH29)[1][2]. This small, hydrophobic protein is rapidly induced by interferons, particularly during viral infections or inflammatory states. It localizes primarily to mitochondria, where it can promote reactive oxygen species (ROS) production, trigger mitochondrial dysfunction, and induce apoptosis[2]. IFI27L2 is upregulated in microglia during aging, ischemic stroke, and pro-inflammatory conditions, where it plays a pivotal role in triggering a pro-inflammatory phenotype and may worsen neuroinflammatory pathology[2]. It can interact with nuclear receptor 4A (NR4A) family members, modulating anti-inflammatory responses by promoting NR4A nuclear export[2]. While strongly implicated in immune defense and brain inflammation, there are currently no drugs known to target IFI27L2 directly, and it is not an established therapeutic target[2][3]. Elevated expression serves as a marker of immune activation, but its use as a diagnostic or prognostic biomarker remains investigational.
