Overview

Interferon alpha-inducible protein 27-like protein 2 (IFI27L2) is a member of the interferon-stimulated gene (ISG) family, closely related to other proteins such as IFI27 and IFI6, and is encoded by the IFI27L2 gene (also known as FAM14A, ISG12B, or TLH29)[1][2]. This small, hydrophobic protein is rapidly induced by interferons, particularly during viral infections or inflammatory states. It localizes primarily to mitochondria, where it can promote reactive oxygen species (ROS) production, trigger mitochondrial dysfunction, and induce apoptosis[2]. IFI27L2 is upregulated in microglia during aging, ischemic stroke, and pro-inflammatory conditions, where it plays a pivotal role in triggering a pro-inflammatory phenotype and may worsen neuroinflammatory pathology[2]. It can interact with nuclear receptor 4A (NR4A) family members, modulating anti-inflammatory responses by promoting NR4A nuclear export[2]. While strongly implicated in immune defense and brain inflammation, there are currently no drugs known to target IFI27L2 directly, and it is not an established therapeutic target[2][3]. Elevated expression serves as a marker of immune activation, but its use as a diagnostic or prognostic biomarker remains investigational.

Mechanism of action

Not directly known as a drug target; no known approved drugs or experimental drugs that modulate IFI27L2 directly Related mechanisms (extrapolated from IFI27 family): induction of apoptosis by mitochondrial pathway, regulation of inflammation through NR4A sequestration

03

Biological functions

Immune response (especially interferon-related, antiviral, and pro-inflammatory signaling)Cell death/apoptosis (via mitochondrial dysfunction and reactive oxygen species generation)Regulation of mitochondrial function (especially in microglia)Interaction with nuclear receptor 4A (NR4A) family to modulate inflammation
04

Disease associations

Inflammation (notably in brain and neuroinflammation)Neurodegenerative disease (role in ischemic stroke pathology and potential roles in neurodegeneration)Infection (response to viral infection as an ISG, potential biomarker for infection severity)
05

Safety considerations

No safety concerns reported for targeting IFI27L2, since it is not a current therapeutic targetTheoretical concerns: interfering with IFI27L2 could affect host immune defense and inflammation regulation
06

Biomarkers

IFI27L2 expression is upregulated in microglia in response to inflammation, aging, and ischemic strokeNot widely used clinically as a biomarker, but its expression correlates with neuroinflammation
